I use both side-by-side, but I think its much more convenient to use this software for not only installing/removing plugins (and avoiding the occasional permissions problems), but also the ability to enable/disable plugins on the fly.
There have also been plugins that, despite my best efforts, just would not load properly through the SIMBL plugins folder.
As much fun as I have rooting through my library sub-directories and running terminal commands, this is a lot less time-consuming.
