On Mac OS X, this updates the WebKit framework that underlies many of the system components such as Dashboard and Safari, and thus should in theory support any other framework that happens to interface with WebKit, including Flash which is bundled with mac OS X, so yes this shouldn't be a problem. Plugins such as Saft or InquisitorX probably will need to be recompiled and slightly rewritten to correctly interface with Safari 4, but in most respects too should be less of an issue than it was between Safari 2 and Safari 3 as WebKit itself has gone through much stabilization and is fairly complete on its backend.New features such as the JavaScript interpreter "Squirrelfish" will introduce notable performance gains, and due to ECMA standardization (Albeit less consistent or industry regarded) will maintain consistent operation. I simply note this because it's obvious that it could be an area of contention for plugins or input loaders which have a direct dependency on said component.
