Existing users, 
Troubles come from the file named vpd.properties in your home folder.
It's a text file, try editing it. Most probably, you moved MystIV folder since it was installed, or renamed one of the enclosing folders. Just correct the various path in this file. It worked for me. Once corrected, the 1.0.1 updater shows the right location.

Dear god, I was wondering what that file was... could they not choose a better place for it? There are, like, folders for that stuff!! WOW! How about ~/Library/Application Support or even ~/Library/Preferences instead of dumping it right into my home folder... where it's likely to get deleted? Lucky I found out what it was before I noticed it anyways and 'DEBTWEETED' it... would have happened sooner or later...Sheesh. At least the game itself is pretty good...
